Title: High throughput evaluation of the payload delivery efficiency for immunoconjugates

Dr. Hou received his PhD degree in 2009 at the Institute of Molecular Biology, Academia Sinica, Taiwan. He then moved to the Genomic Research center, Academia Sinica for postdoctoral training. In 2016, he joined the group of Dr. Sachdev Sidhu as an assistant professor at the Shanghai Advanced Immunochemical Studies (SIAIS), Shanghaitech University, Shanghai. He has over 7 years’ experience in synthetic antibody technology.


The potency of the immunoconjugates depends on the antibody as the guiding modules targeting designated molecules on cell surfaces. The discovery of optimal immunoconjugates is limited by the antibody-payload conjugation procedure. In this work, a cytotoxicity screening system was developed for high throughput discovery or engineering of highly functional payload delivering antibodies. Antibodies with superior delivery efficiency were identified by the combination of this system with phage-displayed synthetic antibody libraries. These results suggest that synthetic antibody libraries built on a consistent framework and unlimited by natural immune system could be a good source for immunoconjugate development. The principles governing the efficiency of the antibodies as targeting modules will also be discussed.
